Året er 1973. Den tidligere elitesoldat James Conrad bliver hyret til at lede en flok eventyrlystne forskere og soldater ud på en ekspedition til den mytiske ø "Skull Island". Formålet er at smide seismiske bomber og kortlægge øens undergrund, men det varer ikke længe inden, at bomberne forstyrrer øens naturlige dyreliv, og så går det pludselig op for ekspeditionens medlemmer, hvilke hemmeligheder Skull Island gemmer på. Her er naturen nemlig gået amok og har frembragt nogle enorme væsner, der tilbedes som guder af de primitive indfødte på øen, og som beskytter deres territorie med sønderrivende effektivitet.
