Lõvi Alex, sebra Marty, jõehobu Gloria ja kaelkirjak Melman püüavad ikka veel pääseda oma armastatud koju, New Yorgi loomaaeda. Loomulikult on nendega kaasas ka ennast kõigi maailmade valitsejaks pidav kuningas Julien, teda kõikjal saatev Maurice ning igast olukorrast võitjana väljuvad pingviinid. Läbi Euroopa rännates leiavad nad endale ideaalse peidupaiga rändtsirkuse näol. Milles tehakse suured Madagaskari stiilis ümberkorraldused!

Tema majesteet Elizabeth II tahab Los Angelese linna külastada. See võrdub loomulikult katastroofiga kui selgub, et vastuvtõtukomitee esimees võib olla narkokaupmees ja mõrvar. Selline juhtum nõuab superpolitseinik Frank Drebini abi. Hoogsalt asub ta sündmuseid uurima ja jätab endast maha kaose raja.
