Helt siden den kommende studenten Mike Wazowski var et lite monster har han drømt om å bli en Skremmer—og han vet bedre enn noen at de beste Skremmerne kommer fra Monsteruniversitetet (MU). Med i det første semesteret på MU, avsporer Mikes planer da han treffer tøffingen James P. Sullivan, “Sulley”, en født Skremmer. Parets helt ville konkurranseånd sørger for at begge blir kastet ut av universitetets Eliteskremmeprogram. Drømmene deres er midlertidig knust, men de innser snart at de må samarbeide, også med en sprø gjeng av mislykkede monstre, hvis de skal klare å lykkes.

Året er 2035, og bare én prosent av jordas befolkning har overlevd et dødelig virus som herjet i 1996. James Cole blir overtalt til å reise tilbake til 1996 for å samle informasjon om hvordan epidemien oppsto, slik at man kan utvikle en vaksine.
